PhD student in the DFG project AQUASHIFT (The impact of climate variability on aquatic ecosystems) within the subproject “Coupling between phyto- and bacterioplankton during early spring bloom conditions”

The focus of my subproject is to investigate the impact of temperature on algae-bacteria coupling and bacterial diversity during early spring bloom conditions. I particularly focused on the phytoplankton bloom phase to investigate how carbon-flow and bacterial community composition is changing with increasing water temperature. I analyze bacterial activity, succession, composition and protist grazing on the bacterial comunity with following methods:

- DNA/RNA extraction, PCR and 16S rRNA cloning

- DGGE

- Sequencing and phylogenetic analysis (ARB)

- Probe design

- CARD-FISH

- MICRO-CARD-FISH

- flow cytometry

- Bacterial production (3H-thymidine and/or 3H-leucine incorporation)

- Primary production (14C-bicarbonate incorporation)

- Microscopy (bacteria, protist)
